LNP characterisation – from LNP formulation to protein corona in human serum

- Typical LNP production process involves nucleic acid(s) mixing with selected lipids, purification and formulation.
- When multiple payloads are used the LNP formulation usually leads to heterogenous particles with more than 90% of impurities.
- Another challenge is selection of proper lipid or modification of the LNP surface to allow for active targeting of the cells.
- Last but not least, when the LNP is delivered via human blood the LNP will be surrounded by proteins (so called corona).
- To better understand all these processes and enable better LNPs design new analytical methods and analytical systems were developed. In this paper the PATfix LNP Switcher and LNP Corona will be presented. These tools are rapidly becoming the gold standard in top pharma and biotech companies working on next-generation LNPs.
